Inverse expression of peroxisomes and connexin-43 in the granulosa cells of the quail follicle.

چکیده
Studying the regulation of peroxisome (Px) expression could improve our understanding of human peroxisomal disorders. The granulosa of the largest preovulatory quail follicles proved to be a relevant model because (a) Px expression changes according to the follicular maturation stage and (b) Px expression varies regionally according to the distance of the granulosa relative to the germinal disc region containing the female gamete (oocyte). The question was asked whether Px expression is related to the extent of metabolic cell coupling and whether zonal Px variation is causally related to oocytal factors. This was evaluated by the presence of catalase and Cx-43 (marker proteins for peroxisomes and gap junctions, respectively) and by in vitro experiments with granulosa explants. The data obtained show that the expression of Cx-43 and Px is inversely correlated both temporally and spatially. Uncoupling of gap junctions results in an upregulation of alpha-catalase immunofluorescence. This is in agreement with reports that gap junctions are often negatively affected by Px proliferators. The zonal gradient in Px expression appears to be imposed by the oocyte, as is the case for steroidogenesis and proliferative capacity in the granulosa epithelium. P-228: Altered Expression of Folliclestimulating Hormone Receptor and Luteinizing Hormone Receptor in Granulosa Cells from Women with Polycystic Ovary Syndrome
Background: Polycystic ovary syndrome (PCOS) is a common complex genetic endocrinopathy, affecting 5 - 10% of women at reproductive age. PCO granulosa cells seem to have abnormal responses to follicle- stimulating hormone (FSH). FSH is considered to be a pituitary glycoprotein that plays an important role during folliculogenesis as it promotes the proliferation and differentiation of granulosa ...
متن کاملEvaluation role of beta-2 adrenoceptor agonist and antagonist in the expression of gap junction genes (connexin 37/43) in cumulus cells of women with poor ovarian response
Background: Connexon is a membrane structural protein in the gap junctions. These cellular connections are responsible for transporting ions and messenger molecules to the oocyte. This study aimed to investigate the role of beta-2-adrenoceptors in the process of follicle growth based on the expression of the two connexins 37/43 in the gap junctions that have a primary role in the mitotic resump...
متن کاملDIFFERENTIATION OF HUMAN OVARIAN FOLLICULAR GRANULOSA CELLS INTO KERATINOCYTES
Background & Aims: Stem cells are undifferentiated cells and are found in different tissues. These cells have capacity of self-renewal and differentiation into other lineages. Granulosa cells (GCs) are the multipotent stem cells. In the present research we evaluated the differentiation potential of GCs into keratinocytes. Material & Methods: GCs were cultured after enzymatic isolation from ova...
متن کاملGap junctions and ovarian folliculogenesis.
Gap junctions are collections of intercellular membrane channels that allow adjacent cells to share small molecules (< 1 kDa). Gap junction channels are composed of connexins, a homologous family of more than 20 proteins. In developing follicles, gap junctions couple the growing oocyte and its surrounding follicle cells into a functional syncytium. This review summarizes evidence on the express...
